At the very least you need to know:
Testosterone (Total, Free, and %Free)
Estrogen E2 Estradiol (Total, Free)
LH (Lutenizing Hormone)
FSH (Follicular Stimulating Hormone)
SHBG (Sex Hormone Binding Globulin)
A Lipids Panel (Cholesterol)
and then there are some others .. that .. might help.. depending on other results. Like:
Prolactin
Progresterone
But at the very least you want a Blood Karotype and FISH. The Buccal Smear is about as inaccurate as it is possible to get. Let me explain.. the Buccal Smear detects "Barr" bodies in the cells. Not very well, nor very accurately. But it is alot less expensive. If you test positive then the Doctor will submit you to other tests .. which if those prove true as well, then we can say with accuracy that the Buccal Smear was correct. Though we only really know that in hindsight. Barr bodies are only present in females and people Intersex (not quite female?). Everyone XXY has barr bodies. Other conditions have barr bodies .. if they're female to some degree.
